Bunlar\u0131n sonuncusu hen\u00fcz 2014\u2019de ger\u00e7ekle\u015fen ve Alvin\u2019in eskisinden y\u00fczde 40 daha fazla hacme sahip yeni bir titanyum k\u00fcreye kavu\u015fmas\u0131n\u0131 sa\u011flayan yenileme. Daha da \u00f6nemlisi, yeni Alvin\u2019in bilimsel \u00f6rnekleme ve yeni sens\u00f6rlerle entegre olma kapasitesi \u00e7ok artm\u0131\u015f durumda. Biyolojik bir \u00f6rnekleme yapt\u0131ysan\u0131z da RNA gibi \u00e7abuk bozulan bir molek\u00fcl\u00fc i\u00e7in daha da \u00f6zel \u00f6rnekleme y\u00f6ntemleri d\u00fc\u015f\u00fcnmeniz gerekir. Ancak in situ olarak kimyasal \u00f6l\u00e7\u00fcmler yapmak bir yere kadar m\u00fcmk\u00fcn, ama derinlik ve dolay\u0131s\u0131yla bas\u0131n\u00e7 artt\u0131k\u00e7a bu da zorla\u015fabiliyor. \u00dcst\u00fcne \u00fcstl\u00fck hidrotermal kaynaklar gibi, s\u0131cak ve kimyasal kompozisyonu \u00e7ok farkl\u0131 olan hidrotermal su ile so\u011fuk dip sular\u0131n\u0131n h\u0131zl\u0131ca kar\u0131\u015ft\u0131\u011f\u0131 ve de\u011fi\u015fkenli\u011fin \u00e7ok fazla oldu\u011fu habitatlar i\u00e7in yerinde ve y\u00fcksek frekansta \u00f6l\u00e7\u00fcm yapmak daha da \u00f6nem kazan\u0131yor.<\/p>\n<p>Asl\u0131nda \u00e7o\u011fu laboratuvar \u00f6l\u00e7\u00fcm tekni\u011fini bas\u0131nca dayan\u0131kl\u0131 bir hazneye koyup in situ bir \u2018sens\u00f6r\u2019 olarak adapte etmek m\u00fcmk\u00fcn. Ancak, derinde \u00e7al\u0131\u015facak bir kimyasal sens\u00f6r i\u00e7in ba\u015fka parametreler de var d\u00fc\u015f\u00fcnecek. \u00d6rne\u011fin, tekni\u011fin y\u00fcksek \u00e7\u00f6z\u00fcn\u00fcrl\u00fckte ve h\u0131zl\u0131 (en fazla saniyeler i\u00e7inde) \u00f6l\u00e7\u00fcm almas\u0131 laz\u0131m. Kalibrasyonlar\u0131n bas\u0131n\u00e7 ve s\u0131cakl\u0131kla \u00f6ng\u00f6r\u00fclebilecek\/geri d\u00f6n\u00fc\u015fl\u00fc \u015fekilde de\u011fi\u015fmesi de gerekli. Bunlara ek olarak, sens\u00f6r sisteminin az g\u00fc\u00e7 harcamas\u0131 ve hacim\/a\u011f\u0131rl\u0131k olarak da minimal olmas\u0131 laz\u0131m \u2013 zira Alvin sepet fotograflar\u0131m\u0131zdan da anlam\u0131\u015f oldu\u011funuz \u00fczere denizalt\u0131n\u0131n a\u015fa\u011f\u0131 g\u00f6t\u00fcrebilece\u011fi \u015feylerin hacmi s\u0131n\u0131rl\u0131.<\/p>\n<p>Biz bu seferde, yukar\u0131daki gerekliliklerin \u00e7o\u011funu kar\u015f\u0131layan elektrokimyasal bir sens\u00f6r seti kullan\u0131yoruz. \u2018Voltammetri\u2019 ad\u0131 verilen bu elektrokimyasal metodta \u00fc\u00e7 iletken elektrot kullan\u0131yor, bunlar\u0131n aras\u0131nda uygulad\u0131\u011f\u0131m\u0131z potansiyel fark sonucu olu\u015fan ak\u0131m e\u011frilerine bak\u0131yoruz. Elde etti\u011fimiz bulgular bize biyolojik topluluklar\u0131n de\u011fi\u015fen s\u0131cakl\u0131k ve oksijen\/hidrojen s\u00fclf\u00fcr kar\u0131\u015f\u0131mlar\u0131na g\u00f6re ni\u015fler i\u015fgal ettiklerini ortaya koyuyor. \u00d6rne\u011fin midyeler (Bathymodiolus thermophilus) bol oksijen ve \u00e7ok az hidrojen s\u00fclf\u00fcr\u00fcn ayn\u0131 anda oldu\u011fu habitatlar\u0131 tercih ederken, hemen yan\u0131ba\u015f\u0131ndaki Riftia ise daha s\u0131cak ve hidrojen s\u00fclf\u00fcr\u00fc bol ortamda bulunuyor. \u00a0Ancak, bu ortamlar \u00e7ok de\u011fi\u015fken. Ge\u00e7mi\u015f y\u0131llara g\u00f6re habitatlarda s\u0131cakl\u0131klar ve hidrojen s\u00fclf\u00fcr seviyelerinin azald\u0131\u011f\u0131n\u0131 farketmeye ba\u015flad\u0131k, bu da midyelerin Riftia habitatlar\u0131n\u0131 ele ge\u00e7irmeye ba\u015flamas\u0131n\u0131 a\u00e7\u0131kl\u0131yor.
